Reduction in rat thymocyte interphase death by calcium depletion.

چکیده
Ca depletion/Interphase death/Rat thymocytes Role of Ca ion in development of radiation damage of rat thymocytes was examined. Free DNA release, an expression of chromatin degradation, and the percentage of erythrosin B stained (dead) cells were increased after incubation of 1000 R X-irradiated thymocytes with normal Ca-containing medium. Incubation of the irradiated cells with Ca ion-depleted medium resulted in marked depression of free DNA release as well as of development of cell death. It was concluded from these results that Ca-dependent process(es) was involved in rat thymocyte interphase death.

Interphase Death of Chinese Hamster Ovary Cells Exposed to Accelerated Heavy Ions
Introduction: Heavy ions are nucleus of elements of iron, argon, carbon and neon that all carry positive electrical charges. For these particles to be useful in radiotherapy they need to accelerated to high energy by more than thousand mega volts. Also the cosmic environment is considered to be a complicated mixture of highly energetic photons and heavy ions such as iron. Therefore, the health ...
متن کاملNitrone spin traps and a nitroxide antioxidant inhibit a common pathway of thymocyte apoptosis.
Oxidative stress has recently been suggested to be a mediator of apoptotic cell death [Buttke and Sandstrom (1994) Immunology Today 15, 7-10], although evidence that this phenomenon is a widespread component of apoptosis is lacking. When rat thymocytes were exposed to the glucocorticoid methylprednisolone (MPS), a progressive increase in intracellular peroxides and a decrease in glutathione (GS...
متن کاملStimulation of calcium-ATPase activity by 3,5,3'-tri-iodothyronine in rat thymocyte plasma membranes. A possible role in the modulation of cellular calcium concentration.
We have previously demonstrated that 3,5,3'-tri-iodo-L-thyronine (T3) produces a very rapid and transient increase in calcium uptake and cytoplasmic free calcium concentration in the rat thymocyte, and have postulated that Ca2+-ATPase may contribute to the overall effect of T3 on cellular calcium metabolism. In the present study, we show that in the rat thymocyte, T3 increased plasma membrane C...
متن کاملElimination of Self-Reactive T Cells in the Thymus: A Timeline for Negative Selection
The elimination of autoreactive T cells occurs via thymocyte apoptosis and removal by thymic phagocytes, but the sequence of events in vivo, and the relationship between thymocyte death and phagocytic clearance, are unknown. Here we address these questions by following a synchronized cohort of thymocytes undergoing negative selection within a three-dimensional thymic tissue environment, from th...
متن کاملStimulation of respiration by mitogens in rat thymocytes is independent of mitochondrial calcium.
The role of calcium in the control of respiration by the mitogen concanavalin A (ConA) was investigated in rat thymocytes. ConA induced an increase in both mitochondrial respiration and the mitochondrial calcium pool. The stimulation of respiration was shown to be independent of the increase in mitochondrial calcium: the calcium pool declined after 3 min, whereas the respiration increase was pe...
